New site-specific recombination systems for genome engineering, VCre/VloxP and SCre/SloxP

What are VCre/VloxP and SCre/SloxP systems?

VCre/VloxP and SCre/SloxP are new site-specific recombination systems for genome engineering.

The Cre/loxP and Flp/FRT systems are gold standard techniques used to perform conditional gene knockouts in mice. Many methods based on these two site-specific recombination systems have been implemented in various organisms. Unfortunately, conflicts arise when using the multiple methods of Cre/loxP and Flp/FRT simultaneously in the same system. For example, due to cross-reactivity, one cannot use more than one Cre/loxP within the same construct.

To address this problem, we have developed two novel site-specific recombination systems: VCre/VloxP and SCre/SloxP. These recombination systems work efficiently in human HEK293 cells and mouse ES cells with no cross-reactivity (references 1 and 2).

The VCre/VloxP and SCre/SloxP recombination systems will encourage novel ideas and concepts for the genome engineering of various organisms, BAC engineering, and biotechnology.
